Yali)采后品质劣变的核心问题,该现象与采收 成熟度、降温方式等因素密切相关。不同降温方式可延缓适时成熟度鸭梨的果心褐变,但其分子调控 机制尚待完善。为探究冷藏期间鸭梨果心褐变的分子机制,以商业成熟度鸭梨为研究对象,采用急速 降温和缓慢降温处理,结合全基因组鉴定和转录组数据,分析DREB基因家族在鸭梨果心褐变过程中 的表达模式。结果发现,从全基因组中共鉴定到157个PbDREB转录因子,基于系统进化分析将其划 分为6个亚家族。基因结构与保守结构域分析表明,同一亚家族成员在外显子-内含子分布模式及 AP2结构域序列特征上具有较高的保守性。启动子顺式作用元件预测显示,PbDREB基因可能受脱落 酸(ABRE)、低温响应(CRT/DRE)等胁迫相关信号调控。转录组数据结合qRT-PCR验证结果表明,9 个候选PbDREB基因在缓慢降温处理下呈现差异性表达且与鸭梨的果心褐变指数呈显著正相关(P< 0.05),推测这些基因可能通过上调的表达水平参与鸭梨果心褐变过程。研究结果为DREB基因家族 在鸭梨采后褐变过程中的功能解析提供了理论依据。
英文摘要:
      Core browning is a key issue in the postharvest quality deterioration of Yali pears (Pyrus bretschneideri Rehd. cv. Yali), and this phenomenon is closely related to the harvest maturity, cooling treatment, and other factors. Different cooling treatments can delay core browning at the optimal harvest maturity, but the underlying molecular regulatory mechanisms remain unclear. To investigate the molecular mechanism of core browning in Yali during cold storage, the commercially mature Yali pears were used as the research object. Rapid and gradual cooling treatments were applied, and genome-wide characterization and transcriptome analyses were performed to study the expression patterns of the DREB gene family during core browning in Yali pears. The results revealed that a total of 157 PbDREB transcription factors were identified from the genome-wide and classified into six subfamilies based on phylogenetic analysis. Analysis of gene structure and conserved domains showed that members of the same subfamily exhibited high conservation in exon-intron distribution patterns and AP2 domain sequence features. Predictions of promoter cis-acting elements indicated that the PbDREB gene may be regulated by stress-related signals, such as abscisic acid (ABRE) and the low-temperature response (CRT/DRE). Combining transcriptome data with qRT-PCR validation revealed that nine candidate PbDREB genes exhibited differential expression and positively correlated with the core browning index (P<0.05) of Yali pears under gradual cooling treatment. It was hypothesized that these genes might be involved in the browning of Yali pears by upregulating gene expression levels. These results provide a theoretical basis for the functional analysis of the DREB gene family in the postharvest browning process of Yali pears.
